Regulation of cargo transfer between ESCRT-0 and ESCRT-I complexes by flotillin-1 during endosomal sorting of ubiquitinated cargo

Ubiquitin-dependent sorting of membrane proteins in endosomes directs them to lysosomal degradation. In the case of receptors such as the epidermal growth factor receptor (EGFR), lysosomal degradation is important for the regulation of downstream signalling. Ubiquitinated proteins are recognised in endosomes by the endosomal sorting complexes required for transport (ESCRT) complexes, which sequentially interact with the ubiquitinated cargo. Although the role of each ESCRT complex in sorting is well established, it is not clear how the cargo is passed on from one ESCRT to the next. We here show that flotillin-1 is required for EGFR degradation, and that it interacts with the subunits of ESCRT-0 and -I complexes (hepatocyte growth factor-regulated tyrosine kinase substrate (Hrs) and Tsg101). Flotillin-1 is required for cargo recognition and sorting by ESCRT-0/Hrs and for its interaction with Tsg101. In addition, flotillin-1 is also required for the sorting of human immunodeficiency virus 1 Gag polyprotein, which mimics ESCRT-0 complex during viral assembly. We propose that flotillin-1 functions in cargo transfer between ESCRT-0 and -I complexes

Activated carbon as catalyst support: precursors, preparation, modification and characterization

    Get PDF
The preparation of activated carbon materials is discussed along selected examples of precursor materials, of available production and modification methods and possible characterization techniques. We evaluate the preparation methods for activated carbon materials with respect to its use as catalyst support and identify important parameters for metal loading. The considered carbon sources include coal, wood, agricultural wastes or biomass as well as ionic liquids, deep eutectic solvents or precursor solutions. The preparation of the activated carbon usually involves pre-treatment steps followed by physical or chemical activation and application dependent modification. In addition, highly porous materials can also be produced by salt templating or ultrasonic spray pyrolysis as well as by microwave irradiation. The resulting activated carbon materials are characterized by a variety of techniques such as SEM, FTIR, nitrogen adsorption, Boehm titrations, adsorption of phenol, methylene blue and iodine, TPD, CHNS/O elemental analysis, EDX, XPS, XRD and TGA

Mobility in a Globalised World 2017

The term mobility has different meanings in the following science disciplines. In economics, mobility is the ability of an individual or a group to improve their eco-nomic status in relation to income and wealth within their lifetime or between gen-erations. In information systems and computer science, mobility is used for the concept of mobile computing, in which a computer is transported by a person dur-ing normal use. Logistics creates by the design of logistics networks the infrastruc-ture for the mobility of people and goods. Electric mobility is one of today’s solu-tions from an engineering perspective to reduce the need of energy resources and environmental impact. Moreover, for urban planning, mobility is the crunch ques-tion about how to optimise the different needs for mobility and how to link differ-ent transportation systems. In this publication we collected the ideas of practitioners, researchers, and gov-ernment officials regarding the different modes of mobility in a globalised world, focusing on both domestic and international issues
